Brown adipose tissue transplantation ameliorates polycystic ovary syndrome.

چکیده
Polycystic ovary syndrome (PCOS), which is characterized by anovulation, hyperandrogenism, and polycystic ovaries, is a complex endocrinopathy. Because the cause of PCOS at the molecular level is largely unknown, there is no cure or specific treatment for PCOS. Here, we show that transplantation of brown adipose tissue (BAT) reversed anovulation, hyperandrogenism, and polycystic ovaries in a dehydroepiandrosterone (DHEA)-induced PCOS rat. BAT transplantation into a PCOS rat significantly stabilized menstrual irregularity and improved systemic insulin sensitivity up to a normal level, which was not shown in a sham-operated or muscle-transplanted PCOS rat. Moreover, BAT transplantation, not sham operation or muscle transplantation, surprisingly improved fertility in PCOS rats. Interestingly, BAT transplantation activated endogenous BAT and thereby increased the circulating level of adiponectin, which plays a prominent role in whole-body energy metabolism and ovarian physiology. Consistent with BAT transplantation, administration of adiponectin protein dramatically rescued DHEA-induced PCOS phenotypes. These results highlight that endogenous BAT activity is closely related to the development of PCOS phenotypes and that BAT activation might be a promising therapeutic option for the treatment of PCOS.

Association between serum levels of Adipokines and Polycystic Ovary Syndrome
Polycystic ovary syndrome (PCOS) is the most common endocrine disorder associated with metabolic disorders and infertility. Evidence suggests that the adipose tissue secreted products, adipokines, play an important role in the pathophysiology of PCOS. Reciprocally, PCOS influences on the adipokines serum content, also on gene or protein expression of some of them. Finding out of assoc...
متن کاملEvaluation of Tnf-α and Il-6 mRNAs expressions in visceral and subcutaneous adipose tissues of polycystic ovarian rats and effects of resveratrol
Objective(s): Some studies suggest that chronic low-grade inflammation is involved in insulin resistance in polycystic ovary syndrome (PCOS). This study assessed possible involvement of alteration in expression of two pro-inflammatory factors, tumor necrosis factor-α (TNF-α) and interleukin-6 (IL-6) in adipose tissues of PCOS rats in the impairment of insulin actions. Also, effects of resveratr...
متن کاملMetabolic and other effects of pioglitazone as an add-on therapy to metformin in the treatment of polycystic ovary syndrome (PCOS).
Insulin resistance is a key pathogenic defect of the clustered metabolic disturbances seen in polycystic ovary syndrome (PCOS). Metformin is an insulin sensitizer acting in the liver and the peripheral tissues that ameliorates the metabolic and reproductive defects in PCOS. In addition, pioglitazone is an insulin sensitizer used in diabetes mellitus type 2 (T2DM), improving insulin resistance (...
متن کاملChanges of The Uterine Tissue in Rats With Polycystic Ovary Syndrome Induced by Estradiol Valerate
Objective : Polycystic ovary syndrome (PCOS) is one of the most common hormonal disorders that can lead to irregular menstrual cycles and hyperandrogenism. Reduced levels of progesterone and increased estrogen in these women can perpetually stimulate the endometrial tissue of the uterus. In this study, the effect of PCOS induction was studied using estradiol valerate. MaterialsAndMethods Adult ...
متن کاملExpression of PPAR-γ in adipose tissue of rats with polycystic ovary syndrome induced by DHEA.
The objective of this study was to investigate the expression of peroxisome proliferator-activated receptor-γ (PPAR-γ) in adipose tissue of the rat model of polycystic ovary syndrome (PCOS), induced by dehydroepiandrosterone (DHEA). Sixteen sexually immature Sprague-Dawley female rats were randomly assigned to the DHEA (n=8) or control (n=8) group. Adipose tissue was collected from the two rat ...
